Upstream process development
Upstream process development is the foundation of biologics manufacturing, shaping how efficiently cells produce a target protein, antibody, or other therapeutic molecule. It focuses on optimizing the biological system before purification begins, with the goal of achieving high yield, consistent quality, and scalable performance.
In a typical upstream workflow, scientists develop and refine the cell line, media, feeding strategy, bioreactor conditions, and process controls. Small changes in pH, temperature, dissolved oxygen, agitation, or nutrient delivery can significantly affect cell growth, productivity, and product quality attributes such as glycosylation, aggregation, and charge variants. Because of this complexity, upstream development relies on iterative experimentation and careful data analysis.
Modern upstream process development is increasingly data-driven. Teams often generate large volumes of experimental results across design-of-experiments studies, screening campaigns, and scale-up runs. Managing this information effectively is critical. Without a structured way to capture protocols, results, and metadata, valuable knowledge can be lost between scientists, programs, or sites. Informatics platforms help centralize data, improve traceability, and make it easier to compare experiments across different stages of development.
Another major challenge is translating results from benchtop systems to pilot and manufacturing scale. A process that performs well in a shake flask or small bioreactor may behave differently when scaled up. Successful upstream development therefore requires robust modeling, standardized workflows, and collaboration between development, analytics, and manufacturing teams. This enables better decision-making and reduces the risk of costly delays later in the lifecycle.
Quality by design principles are also central to upstream development. By identifying critical process parameters and their impact on critical quality attributes early, organizations can build more robust processes and support smoother technology transfer and regulatory readiness.
For organizations developing biologics, the ability to connect experimental data, process knowledge, and team collaboration is essential. Strong upstream process development helps accelerate timelines, improve reproducibility, and increase confidence in moving from research to production.
From DoE studies and screening campaigns to scale-up runs, upstream process development generates the data that determines how efficiently a process performs—and how smoothly it transfers to manufacturing.
